A slide is a glass plate used to place the experimental materials in experiments, which is rectangular in shape, thicker, and has better light transmission; a cover slip is used to cover the material to avoid contact between the liquid and the objective lens, to avoid polluting the lens, which is square in shape, thinner, and has better light transmission.

1. The smear method is a method of preparing a slide by evenly coating the material on the slide glass.
 
The smear materials include unicellular organisms, small algae, blood, bacterial culture liquid, loose tissues of animals and plants, testes, anthers, etc.
 
When applying the coating, pay attention to the following:
(1)The slide must be clean.
(2) The slide must be level.
(3) The coating must be uniform. Apply the liquid droplet to the right side of the slide, and use a surgical blade or a toothpick to evenly coat it.
(4)The coating should be thin. Use another slide as a push slide, and push gently from right to left along the drop of the coating liquid on the slide (the angle between the two slides should be 30°-45°) to form a uniform thin layer.
Fixed. If fixed, it can be done with a chemical fixative or by drying (for bacteria).
(6) Staining. Bacteria are stained with methylene blue, and blood is stained with Wright's stain. The stain liquid should cover the entire smear surface.
Rinse. Use absorbent paper to dry or bake dry.
(8)Sealed Slides. Use Canadian tree resin for long-term preservation.
 
2. The method of pressing the biologic material between a slide and a cover slip, applying a certain pressure to press the tissue cells, is a method of preparing slides.
 
3. Slide preparation is a method of making glass slide specimens by sealing and fixing the biological material as a whole, which can be used to make temporary or permanent slides.
 
The slide materials include: microorganisms such as Chlamydomonas, Spirogyra, Amoeba, and Nematode; Hydra, plant leaf epidermis; insect wings, legs, and mouthparts, and human oral epithelial cells, etc.
 
When making slides using the method, attention should be paid to:
When holding a slide, it should be held level or placed on a platform. The amount of water should be appropriate, enough to cover the cover glass.
(2) The material should be spread out with a dissecting needle or forceps to prevent overlapping, and flattened on the same plane.
(3)Place the cover slip slowly on one side of the droplet to prevent air bubbles.
Step 4: When staining, drop a drop of staining liquid on one side of the cover slide, and use a absorbent paper to attract from the other side to make the specimen under the cover slide uniformly stained. After staining, use the same method, drop a drop of clean water, after absorbing the staining liquid, observe under the microscope.
 
4. Slicing is the preparation of a slide specimen by cutting thin slices from a biological body.
 
Due to different requirements, the tissue can be sliced manually with a blade, or it can be embedded in paraffin or cellulose nitrate or frozen at low temperature, and then sliced with a slicing machine. The tissue is cut into thin slices of 5 to 10 micrometers, which can be observed under an optical microscope. Ultra-thin slices of tissue blocks embedded with epoxy resin or methyl methacrylate have a thickness of 20 to 50 nanometers, which are specially observed under an electron microscope. Generally, the slices of roots and stems used in teaching are called paraffin slices.
