Main Composition
Each reagent bag contains the following reagent components:
(1) Test strip/card (mainly composed of mouse anti-human sperm SP10 protein monoclonal antibody A, mouse anti-human sperm SP10 protein monoclonal antibody B, sheep anti-mouse IgG polyclonal antibody, nitrocellulose membrane, glass fiber, nonwoven paper, absorbent paper, plastic sheet, and tape.): 1 piece/bag;
(2) Desiccant: 1 packet/bag.
2. Reagents not included:
(1) Semen collection cup: 1 per person.
(2) Semen transfer device (or plastic pipette): 1 per person.
(3) Sample dilution tube (with diluent): 1 person/bottle.
Sample Requirements
Sample Collection
(1) Semen should be collected in a dedicated semen collection container or a clean plastic container. The mouth of the bottle should not be too small, otherwise it is easy to miss when collecting semen. The container brought by oneself should be cleaned and dried.
(2) Use manual stimulation (masturbation) or other methods to obtain a semen specimen, and do not use any lubricant or washing liquid; or use a specially designed non-toxic condom for semen collection, as the lubricant on a regular condom may affect the test results.
(3) The collection time for semen sample should be at least 2 days after the last ejaculation and no longer than 7 days. During this time, there should be no ejaculation due to masturbation, nocturnal emission, etc. Short or long collection times can affect the accuracy of the test.
(4) Should collect all ejaculated semen as much as possible, without any omission. If there is any doubt about the test results due to omission, it is recommended to abstain from sexual intercourse for as many days as possible and then collect semen for testing again. Morning ejaculation is the best time for collection. The method of interruption of ejaculation during sexual intercourse cannot be used for collecting semen, because the initial part of the ejaculation may be lost, and this part of the sperm concentration is often the highest. At the same time, it is inevitable to mix with vaginal secretions, which may affect the accuracy of the test results.
(5) Should as much as possible use fresh semen samples for detection (recommended for detection within 3 hours), if you need to try again later, the semen sample (sealed) must be stored at 2~8℃ for no more than 12 hours. Longer-term storage should be at or below -20℃, but it can only be dissolved once.
Sample Preprocessing
(1) After semen is ejected into the collection container, it quickly forms typical semi-solid gel-like clumps. It should be left at room temperature for 30 to 60 minutes, and it will completely liquefy (i.e., become homogeneous and more watery, with possible gel-like clumps that do not liquefy being normal). It is rare for it to take more than 60 minutes or longer. If it has not completely liquefied after 60 minutes, it should be considered abnormal. It is recommended to take another sample for observation two days later. If it still cannot completely liquefy after 60 minutes, it should consult a specialist doctor.
Only a fully liquid semen sample can be used with this reagent for testing.
The liquefied semen sample must be thoroughly mixed within its original container, but it should not be agitated violently.
(2) Use a semen transfer device to absorb 0.1 ml of completely liquid semen and add it to a small tube containing sample diluent (composition: 0.1M phosphate-buffered saline (PBS) (pH 7.4±0.2) + 1% Tween-20) (1.9 ml per tube). Alternatively, you can use a disposable plastic pipette that comes with the kit to absorb the semen specimen and drop 3 drops (35 microliters) into the small tube containing the sample diluent.
Remark: Avoid absorbing the gel-like or viscous substance in semen.
(3) Tighten the cap of the sample dilution tube, thoroughly mix the semen and the sample diluent, that is, invert and rotate the sample processing tube at least 5-10 times. If the sample is very concentrated or very viscous, it should be inverted and rotated another 10 times, or directly stirred for 10-20 seconds. Finally, leave the small tube containing the semen to stand for 2 minutes.
