significance of detecting sperm density

according 【who manual of human semen laboratory examination (5th edition) mentioned that the main factors affecting male infertility include: semen volume, total sperm count, sperm concentration, liquefaction time, semen ph value, sperm normal morphology rate, among which sperm concentration detection is an important indicator to measure male infertility. in this manual, the normal minimum value of sperm concentration is 15x 10.⁶piece/ml, when the sperm concentration is less than 15x10⁶piecewhen/ml, it is considered that there is fertility difficulty. however, male infertility needs to be combined with other indicators to make a final judgment. based on several anatomical, genetic and biochemical characteristics, sp10 protein has been proved to be an analyte of sperm concentration, which is only expressed in testes and is the differentiation marker of the last step in the process of spermatogenesis. according to the research data, sp10 protein concentration has a certain linear relationship with sperm concentration, that is, when sperm concentration ≥ 15x10⁶pieceat/ml, sp10 protein test showed positive. therefore, this reagent can be used to assist the auxiliary diagnosis of male infertility and provide birth guidance for suitable-age couples.

(1) test strips/cards (mainly composed of mouse anti-human sperm sp10 protein monoclonal antibody a, mouse anti-human sperm sp10 protein monoclonal antibody b, sheep anti-mouse igg monoclonal antibody, nitric acid fiber film, glass fiber, non-woven paper, absorbent paper towels, plastic sheet, tape composition.) : 1 piece/bag;

(2) desiccant: 1 pack/bag.

2. reagent components required but not included:

(1) semen collection cup: 1/person portion;

(2) semen transferer (or plastic dropper):1 piece/human portion;

(3) sample dilution tube (including diluent):1 person/bottle.

[sample requirements]]

sample collection

(1) semen should be collected in a special semen collection container or a clean plastic container. the bottle mouth should not be too small, otherwise, it is easy to miss when collecting semen. the self-provided container should be cleaned and dried.

(2) use artificial stimulation (masturbation) or other methods to obtain semen samples, and do not use any lubricating fluid or lotion; or use non-toxic condoms specially designed for semen collection for semen collection, because lubricating fluid on ordinary condoms may affect the test results.

(3) the collection time of semen samples should be at least 2 days after the last ejaculation and no more than 7 days after the last ejaculation, and there should be no cases of masturbation, spermatorrhea and so on during this period. too short or too long will affect the accuracy of the test.

(4) collect all the injected semen as much as possible and must not be missed. if there are indeed omissions and doubts about the test results, the semen should be collected for testing in the same number of days as possible for abstinence again, and it is best to sperm collecting in the morning. interruption of sexual intercourse and sperm discharge cannot be used to collect semen, because the initial part of ejaculation may be lost, and this part of sperm concentration is often the highest. at the same time, it is inevitable to mix into vaginal secretions and affect the accuracy of the test results.

(5) fresh semen samples should be used for testing as much as possible (testing within 3 hours is recommended). if you want to try again later, semen samples (sealed) should be stored at 2 ~ 8℃, but the storage time should not exceed 12 hours. longer storage should be at-20℃ and below, but it can only be dissolved once.

2. sample pretreatment

(1) the typical semi-solid gel mass will appear soon after the semen is injected into the collection container. generally, it should be placed at room temperature for 30~60 minutes, and it will completely liquefy (that is, it becomes homogeneous and thinner, it is normal that it may contain non-liquefied gel-like lumps), rarely exceeding 60 minutes or longer. if it is not fully liquefied for 60 minutes, it should be regarded as abnormal. it is recommended to take another sample for observation after 2 days. if it is still not fully liquefied for 60 minutes, consult a specialist.

only completely liquefied semen samples can be tested with this reagent.

the liquefied semen sample must be fully mixed in the original container, but cannot be shaken violently.

(2) use the semen transfer device to absorb 0.1 ml of completely liquefied semen and add it to diluent containing samples (ingredients: 0.1m phosphate buffer pbs(ph 7.4±0.2)+ 1% tween-20) small tube (1.9 ml/tube), or use a disposable plastic dropper matched with the kit to absorb semen samples, and add 3 drops (35 microliters) to diluent containing samples small tube.

note: avoid absorbing colloidal or viscous substances in semen.

(3) close the bottle cap of the sample dilution tube, mix the semen and sample diluent thoroughly, and rotate the sample processing tube upside down at least 5-10 times. if the sample is very thick or very viscous, it should be rotated upside down for 10 times again, or stir directly for 10-20 seconds, and finally leave the small tube containing semen for 2 minutes.
